Y-DNA and mtDNA are particularly useful in genealogy because they follow two specific lines through a family tree. Y-DNA follows the direct paternal line from father to son, while mtDNA follows the direct maternal line from mother to child.
Recording these results alongside the people tested, their ancestral lines, and their earliest known ancestors can make it much easier to compare results and preserve your DNA research over time.
Y-DNA vs. mtDNA: What Does Each Test Trace?

Y-DNA is passed from father to son and can therefore be used to investigate a person's direct paternal line. It can be especially useful when comparing men who descend from the same paternal lineage, including surname-based research.
โ mtDNA โ Direct Maternal Line
Mitochondrial DNA is passed from a mother to her children. Both males and females inherit mtDNA, but only females pass it on to the next generation. This makes mtDNA useful for investigating the direct maternal line.

Important: A Haplogroup Does Not Identify One Specific Ancestor
A haplogroup represents a branch of a much larger genetic family tree. Sharing a Y-DNA or mtDNA haplogroup does not, by itself, prove that two people descend from the same recent ancestor.
More specific subclades can sometimes provide additional information about how closely related two paternal or maternal lines may be, but the appropriate interpretation depends on the test resolution, the specific haplogroup, and the available genealogical evidence.
Geographic associations and migration patterns should also be treated as research clues rather than proof that a particular ancestor lived in a specific place.

Step-by-Step Haplogroup Research Workflow
- Identify the direct line. Determine whether you are researching the direct paternal Y-DNA line or the direct maternal mtDNA line.
- Record the tested relative. Enter the person's name, relationship, testing company, and relevant test type or level.
- Record the genetic result. Enter the assigned haplogroup, subclade, terminal SNP, or relevant mtDNA information.

Can haplogroups prove where my ancestors lived?
Not by themselves. Haplogroup distributions and geographic associations can provide useful historical clues, but establishing where a specific ancestor lived requires additional genetic, historical, and genealogical evidence.
Should I update the spreadsheet when a haplogroup changes?
Yes. Haplogroup assignments can become more specific as additional testing and new branches are identified. Keeping the spreadsheet updated helps preserve the development of your research over time.
